First Report of Colletotrichum fructicola Causing anthracnose on macadamia in China.

Macadamia (Macadamia ternifolia Maiden and Betche) is an important commercial crop in the world and has the reputation of being the king of nuts (Liu et al. 2019). In August 2020, symptoms of anthracnose appeared on leaves of macadamia in Chongzuo, Guangxi Province, China, with an incidence of 15-20%. The disease developed from the edge of leaf. Initially, the disease symptoms on leaves were faded green spots, light yellow. After expanding and linking together, the leaves appeared brown or black irregular spots, and the edges of diseased leaves dried up and formed large necrosis, eventually leading to defoliation. A large number of orange-yellow spots (acervuli) developed on the diseased parts. Under high humidity conditions, the diseased part was grayish-brown or black, and a large number of yellowish-brown conidia were produced on the leaf surface (Fig.1 A-E). Ten symptomatic leaves were collected and washed with distilled water. Twelve lesion marginal tissues were sterilized with 75% ethanol (V/V) for 30 s and 1% NaOCl for 1min and rinsed with sterile distilled water, plated on potato dextrose agar (PDA) and incubated at 28°C under light. After 3 days, the incubated samples all produced similar cultural morphology. One isolate named GXMC2 as a representative was selected for following study. The colony by single-spore purification on PDA were grayish green with a white outer ring and cottony on surface, pale blackish green in reverse side (Fig.1 F). Conidia with oil droplets were solitary, cylindrical, transparent and measuring 13.78 to 19.25 μm (average 16.90 μm) × 5.14 to 7.33 μm (average 6.23 μm) (n=100) (Fig.1 G). Appressoria were brown to dark brown, with different shapes such as ovoid, elliptical or irregular, some with lobes. The average size was 7.89 to 13.25 μm (average 10.64 μm) × 5.76 to 9.02 μm (average 7.86 μm) (n=100) (Fig.1 H). No setae were found. The isolate was identified as Colletotrichum fructicola on the basis of the morphology of the colonies, conidia and appressoria (Park et al. 2018). The six target gene sequences, including internal transcribed spacer (ITS), β-tubulin (TUB), actin (ACT), histone3 (HIS3), chitin synthase A (CHS), and glyceraldehydes-3-phosphate dehydrogenase (GAPHD) (Qiu et al. 2020), were selected for PCR amplification. The resulting sequences were deposited in GenBank under accession numbers MZ821661, MZ821660, MZ821662, MZ821663, MZ821664 and MZ821665 respectively. Phylogenetic analysis of the concatenated sequences was performed with MEGA 7.0 software. The isolate was grouped in the same clade as other C. fructicola (Fig.2). In May 2022, Inoculation was conducted in the field. Four-year-old macadamia leaves were disinfected with 75% alcohol. The conidial suspension was sprayed on 5 unwounded healthy leaves, and 5 leaves sprayed with sterile distilled water served as control. The experiment was replicated 3 times, with each replicate containing 5 leaves. The average daily temperature and average daily relative humidity in the field were 30°C and 62%, respectively. After 2 days, yellow-brown spots appeared on the inoculated leaves and expanded outward. After 4 days, the diseased areas were dark brown, and the controls remained asymptomatic. The same fungal pathogen was reisolated and purified from inoculated leaves and the identity was confirmed by morphological characterization and molecular analysis, confirming Koch's postulates (Fig.1 I-J). In China, C. fructicola has been reported on Passiflora edulis Sims, Brassica parachinensis, Illicium verum, Peucedanum praeruptorum, etc. (Li et al. 2021; Yu et al. 2022; Zhao et al. 2021; Ma et al. 2020). To our knowledge, this is the first report of anthracnose of macadamia caused by C. fructicola in China. The effect of flavonols in Anacardium occidentale L. leaf extracts on skin pathogenic microorganisms.

Cashew (Anacardium occidentale L.) leaf is traditionally used to treat skin infections. Although many flavonols have been identified from its leaf extract, their inhibitory effects on skin pathogens are not yet determined. The aims of this study were to determine the antimicrobial (against skin pathogenic microbes) and antioxidant activities of four flavonol glycosides from the crude extract and three flavonol aglycones from the hydrolyzed extract. The hydrolyzed extract was found to show higher activities than the crude extract. Myricetin showed the highest activity against all the tested bacteria and yeast with the lowest Minimum Inhibition Concentration (MIC) of 7.81 μg/mL on Corynebacterium minutissimum ATCC23348. Myricetin also exhibited good primary antioxidant activities with the effective concentration with 50% of activity (EC50) values ranged between 2.23 μg/mL and 6.40 μg/mL. The highest secondary antioxidant activity was indicated by myricetin-3-O-rhamnoside. Vacuum packaging improved inactivation efficacy of moderate dry heat for decontamination of Salmonella on almond kernels.

The goal of this study was to develop dry heat processing conditions that could achieve a >5-log reduction of Salmonella with minimal negative impact on almond quality. The effects of almond's water activity (aw) levels and packaging methods on Salmonella inactivation by dry heat were determined. Almonds were dip-inoculated in a four-strain Salmonella cocktail and conditioned to aw of 0.43, 0.33, 0.23, and 0.20. The inoculated almonds were then placed in vacuum-sealed mylar bags (vacuum packaging), ambient-sealed glass tubes (non-vacuum packaging), and petri dishes without covers (no packaging). The packaged and un-packaged almonds were treated by dry heat with 13 % relative humidity at 73 °C. Vacuum packaging in general achieved slightly better (in some cases significantly better (p < 0.05)) or similar inactivation effect on Salmonella than non-vacuum packaging. Both vacuum and non-vacuum packaging methods achieved much greater Salmonella inactivation than the no packaging method. For example, a 4-h treatment at 73 °C reduced Salmonella on almonds with aw of 0.43 by 5.1-, 4.4-, and 1.3-log for mylar bag, tube, and petri dish, respectively. Higher aw levels resulted in better inactivation of Salmonella. To achieve a >4-log reduction of Salmonella on almonds packaged in mylar bags, 3-, 6-, 8-, and 8-h of heat treatment were needed for almonds with aw values of 0.43, 0.33, 0.23 and 0.20, respectively. Vacuum packaging in combination with a 4-h heat treatment of almonds with initial aw of 0.43 or 8-h heat treatment of almonds with initial aw of 0.33 could achieved a ≥5-log reduction of Salmonella. Those two combinations resulted in very little weight loss (≤0.05 %), insignificant color change (∆E ≤ 1.26), and unnoticeable change in visual appearance of almonds, demonstrating that they could be potentially used for raw almond pasteurization. https://doi.org/10.1016/j.ijfoodmicro.2022.109849


Engineered pine nut shell derived activated carbons for improved removal of recalcitrant pharmaceuticals in urban wastewater treatment.

Novel powdered activated carbons (PACs) from pine cones and pine nut shells (PNSs) were tested for the competitive adsorption of pharmaceutical compounds (PhCs) in spiked (100 µg/L) secondary effluent and mixed liquor from an urban wastewater treatment plant. Steam activated PNS77, with hierarchical pore structure and 1463 m2/g of BET area, outperformed the commercial benchmark (WP220, mineral origin) for PhCs and dissolved organic matter (DOM) control. PNS77 attained the highest adsorption capacity and rate in synthetic and real wastewaters. Competitive adsorption isotherms revealed the detrimental effect of direct site competing DOM on PhC removal. The PhCs' adsorbability increased with their hydrophobicity, regardless of the water matrix. Kinetic data allowed inferring that indirect competition due to pore constriction/blockage appeared to occur only in mixed liquor. Adsorption isotherm data modelling for ng/L range revealed 80 % removal of carbamazepine and diclofenac would be achieved dosing 8-15 mg/L PNS77 to secondary effluent, while for mixed liquor the dose must be doubled to balance the increased competition. Hydrophilic sulfamethoxazole required a higher dose (34-44 mg/L), lower in the mixed liquor. Global systematic review and meta-analysis on prevalence and concentration of aflatoxins in peanuts oil and probabilistic risk assessment.

Exposure to mycotoxins in food is largely unavoidable, and concerns about their health effects are growing. Consumption of vegetable oils such as peanuts oil has increased, hence several studies have been conducted on concentration of aflatoxins (AFs) in peanuts oil. Search was performed in Scopus and PubMed databases on prevalence and concentration of AFs in peanuts oil from 1 January 2005 to 15 April 29, 2022. Prevalence and concentration of AFs in peanuts oil was meta-analyzed based on country and type of AFs subgroups. In addition, health risk was calculated using monte carlo simulation method. Pooled prevalence of AFB1 in peanuts oil was 47.9%; AFB2, 46.45%; AFG1, 46.92% and AFG2, 54.01%. The Overall prevalence of AFTs was 49.30%, 95%CI (35.80-62.84%). Pooled concentration of AFB1 in peanuts oil was 2.30 μg/kg; AFB2, 0.77 μg/kg; AFG1, 0.07 μg/kg; AFG1, 0.28 μg/kg. The sort of country based on mean of MOEs in the adults consumers was Japan (47,059) > China (17,670) > Ethiopia (7,398) > Sudan (6,974) > USA (1,012) and sort of country based on mean of MOEs in the children was Japan (120,994) > China (46,991) > Ethiopia (19,251) > Sudan (18,200) > USA (2,620). Hazelnut peptide fractions preserve their bioactivities beyond industrial manufacture and simulated digestion of hazelnut cocoa cream.

Cold press hazelnut cakes represent a concentrated source of proteins that can be industrially exploited. Previously, bioactive attributes of hazelnut protein hydrolysates including antihypertensive and antidiabetic activities were documented. Here, we made an attempt to utilize bioactive hazelnut protein hydrolysates (1 % w/w) in the manufacture of industrial hazelnut cocoa cream and investigate their stability through processing and simulated gastrointestinal digestion. The inclusion of bioactive peptide fractions was a safe practice in the microbiological sense. Proteolysis lowered the potential allergenicity of hazelnut proteins in the cocoa cream products up to about 20 %. In silico trypsinolysis predicted partial degradation for 51.8 % of the peptide sequences (i.e., 43/83) that were present in the hydrolysates. However, partial degradation and mixing of degraded vs non-degraded peptides preserved and/or further elevated bioactive attributes in the digested cocoa cream products in terms of Angiotensin converting enzyme (ACE)-inhibitory (up to about 92 %) and antidiabetic activities (between 7.5 and 44.4 %). In most cases, however, antioxidative activity was < 10 %. While simulated in vitro digestion potentially influenced the bioactive attributes of protein hydrolysates, the influence of cocoa cream processing and food matrix were relatively limited for hydrolysate fractions and more pronounced for protein isolates. Use of Hazelnut Perisperm as an Antioxidant for Production of Sustainable Biodegradable Active Films.

Utilization of food-waste-derived bioactive compounds with biodegradable polymers is an attractive strategy leading innovation in the food packaging sector and contributing to reduce the environmental concerns of plastic packaging disposal. In this field, this work is aimed to use hazelnut perisperm as an antioxidant agent in the production of biodegradable polymeric films for active packaging applications. For this purpose, hazelnut perisperm of a selected particle size (&lt;250 μm) at different percentages (0%, 5% and 10% by weight) was added to a bioderived and compostable polymer suitable for food contact, known as Ecovio®. The blends were produced by a twin-screw extrusion process, while active films were prepared with a pilot lab-scale film blowing plant. The films were characterized in terms of physical-mechanical properties (thermal, tensile, oxygen barrier, optical, sealing ability) and antioxidant activity (DPPH), to investigate their potential use as active packaging. The results showed that the presence of the hazelnut perisperm confers significant antioxidant activity to the films, which is useful in counteracting lipid oxidation and preserve the quality of lipophilic foods, e.g., nut-dried fruits. An extension of the sealability temperature range of the films without compromising their strength was also highlighted. Moreover, the hazelnut perisperm causes a gradual decrease in the stiffness and mechanical strength of the films and an increase in the ductility of the system. https://doi.org/10.3390/polym14194156


Developing sustainable land-use patterns at watershed scale using nexus of soil, water, energy, and food.

Aside from the many services that soil provides, it also stores water and makes it available to crops, which is critical for food security. However, the necessity of further studies for overcoming the existing gap in relation to the role of soil in the water, energy, and food nexus system has been preoccupying the experts and specialists around the world for some time. In this sense, the balance between many key ecosystem components based on the Soil, water, energy, and food (SWEF) nexus framework is one of the key characteristics of holistic and accommodative watershed management systems. To the best of our knowledge, the watershed scale is used as a planning unit for the first time in the current study to construct a conceptual model for adaptive management of optimum land-use/cover allocation using SWEF. The method was then used for the Shazand Watershed, Iran. Numerous metrics, such as soil erosion, soil organic carbon (SOC), water and energy use, mass efficiency, and economic efficiency, were investigated. Finally, a compound indicator was used to generate the SWEF nexus index (SWEFNI) for various land-uses/crops for the node year 2014. SWFENI ranged from 0.19 (worst) for rangeland to 0.78 (best) for almond plantations, according to the findings. The effect of nut growth limitation on triose phosphate utilization and down-regulation of photosynthesis in almond.

There is a controversy regarding when it is appropriate to apply the irrigation restriction in almond trees (Prunus dulcis Mill.) to save water without penalizing yield. We hypothesized that knowing when plants demand fewer photoassimilates would be a good indicator of less sensitivity of the crop to water deficit. One parameter that defines the photosynthetic capacity is the triose phosphate utilization (TPU). Due to its connection to the export of sugars from the leaves to other sink organs, it is a good candidate for being such an indicator. The objective was to analyze the seasonal evolution of the photosynthetic capacity of three almond cultivars (cvs. Guara, Marta and Lauranne) subjected to water stress during vegetative, kernel-filling, and postharvest stages. Two sustained deficit irrigation (SDI) treatments (SDI75 and SDI65 with water reductions of 25% and 35%, respectively) and a control treatment (FI) consisting on full irrigated trees were applied. The response of curves AN-Ci was analyzed to assess the maximum carboxylation rate (Vcmax), maximum rate of electron transport (Jmax), TPU and mesophyll conductance to CO2. Additionally, leaf water potential and yield were measured. Our experimental findings showed any significant differences in the variables analyzed among cultivars and irrigation treatments. However, consistent differences arose when the results were compared among the phenological stages. During the kernel-filling and the postharvest stages, a progressive limitation by TPU was measured, suggesting that the demand for photoassimilates by the plant was reduced. This result was supported by the correlation found between TPU and fruit growth rate. As a consequence, a downregulation in Jmax and Vcmax was also measured. This study confirms that the kernel-filling stage might be a good time to apply a reduction in the irrigation and suggests a method to detect the best moments to apply a regulated deficit irrigation in almond trees. https://doi.org/10.1093/treephys/tpac122


Grazing with trees – A silvopastoral approach to managing and restoring drylands

Trees in dryland forests and wooded areas provide key ecosystem services such as animal feed, timber, fruits and, regulation of soil and water cycles. Equally, the presence of livestock in dryland woody areas can also play an important role in the local ecosystem; not only are they a source of income for local communities, but they also help vegetation and mobilize stored biomass. When both of these ecosystem elements are wisely combined – livestock and trees – it creates an integrated agricultural system that can boost the local ecosystem, representing a welcome agro-ecological transition in livestock farming. The ‘Grazing with Trees’ report gives a thorough assessment of the positive role that optimized extensive grazing livestock farming can play in the management and restoration of drylands’ forests and lands with trees. It assesses and provides sound evidence on the benefits of applying an integrated landscape approach and utilizing farmers and pastoralists’ knowledge to halt desertification, increase resilience, and enhance food security under the actual changing scenario.
